I purchased the intank evo 13.5 placing it in chamber 1 with poly fill on top chemo blue in middle wanted to see what I should place in bottom of intank? 2nd chamber I am placing my protein skimmer and under I have bio max media in a bag . Last chamber is my heater ato and return pump with I’ve sterilizer. What media would be suggested to add to the bottom . I have a mixed reef with 2 clowns and a 6 stripe. Thank you

I think you have it covered, but you could put a piece of sponge in and it will act as both mechanical filtration and bio. You could put a little bag of bio-balls in.

I also seen a video someone takes the original foam filter and cuts it in half to use the only thing is I heard that it is bad to use because it almost impossible to clean

You're using ChemBlue already, so one or the other. I would not use both in a newer smaller tank.

Sponges are easy to rinse IME. I haven't used poly fill or floss in decades.
