encystment
blank.gif

To form or become enclosed in a cyst (resting spore). There are several types of encystment. Stressed cells can 'round up' and settle out of the water column and yet be viable if the stress conditions are removed. Others involve temporary cysts for asexual reproduction, flotation, and other functional aspects of individual survival. Yet another type of encystment involves sexual reproduction and the production of thick-walled hypnozygotes which can remain encysted for months, even years (Steidinger and Tangen, 1996).

life cycle
blank.gif

A continuum of phases and cell types in the reproduction and growth of a species. The life cycle usually contains at least an asexual phase in which a cell can divide by binary fissionand produce two similar cells (1N). It may also contain a sexual phase in which gametesfuse to form zygotes (2N) and these zygotes produce 1N cells.

excystment

When the hypnozygote matures and is ready to produce a motile cell from the resting cell, a naked cell will emerge from an opening in the cyst wall. This emerging cell will either be flagellated or amoeboid. Typically, this cell will undergo meiosis and produce four vegetative 1N cells that are motile in the water column (Steidinger and Tangen, 1996).

sexual reproduction

A method of reproduction that involves two gametes (1N) that fuse to produce a zygote(2N)

Run down Update:
Prep
  • Dumped in enough food to cause a massive outbreak of cyano and dinos that would normally last at least 1-2 months
Before Dosing
  • Turned off Automatic Water Changes
  • Took out GAC (would have taken out any GFO too if I had any)
  • No ozone (same would apply to UV or dosing H2O2)
  • Cut carbon dosing in half
  • Tuned down the skimmer but kept it aerating the water
  • Kept ATS online
  • Kept ATO online
  • Kept feeding as normal (a lot)

First 3 days
  • Last dose last night. Dosed 5 grams per day for 3 days for around 200 gallon system volume. (This works out to around 250mg per 10 gallons per day for 3 days.)
End of day 3 going into day 4
  • Added filter sock.(100 micron felt. 4" ring by 14" length) Also had to change that sock already
  • Blew of all surfaces
  • Brought skimmer back online
Days 4 - 5
  • Changing filter sock 3 times per day
  • Blowing off all surfaces once per day in the evening
  • Minor grow back of dinos each day
  • extremely minor to no growth back of cyano each day

Minor update on dosing for 4 more days.

OK I have been discussing this with my wife.
So between me and her we have come to the conclusion to dose for 7 days.
I will dose for 7 days starting today.

I personally would hold off for 1 week as I run this 7 day plan.
I do not want any harm to come to other members tanks if they start dosing 7 days before I have results.
You can run another 3 day dose as we know there is no harm with this but I can not promise any conclusive outcome of the dinoflagellates cystic state.

Tonight I'm just at 48 hours from my last dose. Since I'm seeing encysted cells now I'm thinking of doing 4 more days.

Thoughts?

If you are willing to work with this hypothesis yes I would keep dosing.
So we know now that they will evolve into a cystic state. Now the question is can we penetrate this wall with continued dosing.
I have done my third dose and it is visible that reproduction comes to a standstill still.
I will have some shots some time today.

Day 6
Much much fewer noticeable dino bubbles growth tonight. I had to "zoom" in to see the little amounts of bubbles so they are kind of blurry.

Ok so today yesterday was dose 5. I had dinos but i believe they were halted in reproduction. But when i did the bucket test i had some that sank and some that floated in the middle of the bucket hence I'm thinking some cells were dead but some were alive, and i had some on top. So what i am thinking is that the cysts had just hatched per say amd hasn't gotten the full effect of the metro. I did a 5 gallon water change, siphoned out most of the dinos that i could. A lot of gino was still left in the water column. Im going the skimmer will take care of a majority of it. After the water change i decided to up the dose to 156 mg per 10 gallons. (One spoon more) now to wait and keep an eye on the tank. My rainbow bubble tip has split in the last two days. My water chemistry is perfect except the nitrates amd small amount of phosphates. Everything else is growing sooooo fast its unreal so it makes me believe that it split due to perfect anemone conditions! I dont think metro had an impact on the split though.

I didn't see a single mobile or recognizable peridinium dinoflagelets tonight in this sample. I took it from the same spot up top on a rock by my RBTAs as the day 3 sample. Found a bubble and sucked it up.

Saw some what I think are encysted ones. And some of those littler odd ball ones that don't move so well. Some kind of phytoplankton.

Pictures and vids uploading now. Will get them posted here tomorrow

Edit: I guess later today.
